Brassica Expression Profiling

Overview
Cogenics, the leading gene expression profiling service provider, offers the first microarray-based Brassica expression profiling service. The microarray was designed in collaboration with the John Innes Centre and the J. Craig Venter Institute and enables researchers to explore gene expression patterns in all Brassica species.  The EST assemblies represented on the microarray are included in the annotation of the emerging Brassica rapa genome sequence (http://brassica.bbsrc.ac.uk/).

Array Specification
  • 91,854 60-mer oligonucleotide probes derived from assemblies of 803,326 public ESTs
  • For use with all Brassica Species
  • Probes are synthesized in situ using Agilent Technologies’ SurePrintTM technology in 2 X 105K format
Cogenics’ Brassica Expression Service Features & Benefits
  • Expert assistance with experimental design
  • Extensive experience in RNA isolation
  • Service provides expression profile of the Brassica genome performed by an Agilent Certified Service Provider
  • Custom bioinformatics and data analysis

Validation
Experiment:
Four biological replicate samples derived from the leaves of B. oleracea (Alboglabra A12DH), B. rapa (Trilocularis RO18) and two reciprocal resynthesized B. napus lines were profiled using this microarray.

Results:
  • A principal component analysis of all the expression data revealed a clear separation of each of the biological replicates based on species (Figure 1).
  • An evaluation of the probes that were significantly detected in all replicates of each genotype revealed genome-specific expression (Figure 2).
